电话:400-6721-600 Human sCD163 ELISA Kit DESCRIPTION CD163 is a protein that in humans is encoded by the CD163 gene. It belongs to the scavenger receptor cysteine rich family type B and consists of a

1048 amino acid residues extracellular domain, a single transmembrane segment and a cytoplasmic tail with several splice variants. It is a scavenger receptor for the hemoglobin-haptoglobin complex. It has also been shown to mark cells of monocyte/macrophage lineage. A soluble form of the receptor exists in plasma, commonly denoted sCD163. It is generated by ectodomain shedding of the membrane bound receptor. sCD163 is upregulated in a large range of inflammatory diseases including liver cirrhosis, type

2 diabetes, macrophage activation syndrome, Gaucher'

s disease, sepsis, HIV infection, rheumatoid arthritis and Hodgkin Lymphoma. PRINCIPLE OF THE ASSAY This assay employs the quantitative sandwich enzyme immunoassay technique. A monoclonal antibody specific for human sCD163 has been pre-coated onto a microplate. Standard, samples and biotin-linked detect antibody specific for sCD163 are pipetted into the wells and sCD163 present is bound by the immobilized antibody and detect antibody following incubation. After washing away any unbound substances, streptavidin-HRP is added. After washing, substrate solution is added to the wells and color develops in proportion to the amount of sCD163 bound in the initial step. The color development is stopped and the intensity of the color is measured. LIMITATIONS OF THE KIT FOR RESEARCH USE ONLY. NOT FOR USE IN DIAGNOSTIC PROCEDURES. The kit should not be used beyond the expiration date on the kit label. Do not mix or substitute reagents with those from other lots or sources. If samples generate values higher than the highest standard, dilute the samples with Assay Buffer and repeat the assay. If cell culture supernate samples require large dilution, perform an intermediate dilution in culture medium. Any variation in standard dilution, operator, pipetting technique, washing techniques, incubation time or temperature, and kit age can cause variation in binding. This assay is designed to eliminate interference by factors present in biological samples.
